Syros Pharmaceuticals, Inc. is a biopharmaceutical company with a pipeline that includes three clinical-stage assets: SY-2101, SY-5609 and tamibarotene.

SY-2101 Overview

SY-2101, a novel, oral formulation of arsenic trioxide (ATO), offers a unique opportunity to build off the established efficacy of intravenously administered ATO in newly diagnosed acute promyelocytic leukemia (APL) while significantly improving the lives of patients by reducing treatment burden of the IV ATO regimen, which involves up to 140 infusions over a typical 10-month course.

Key Strengths

If approved, SY-2101 could become the new standard of care for APL treatment, potentially expanding into other malignancies where ATO has shown promise. Key attributes include:

  • Phase 3-Ready Asset: FDA- and EMA-aligned path to approval in frontline APL and potential to leverage Phase 1b data for a streamlined pivotal study. The trial assessed a Phase 3 dose, food effect, and safety of extended administration
  • Orphan Drug Designation: Granted in US and EU, providing meaningful exclusivity periods
  • Novel Oral Formulation: Of an approved, efficacious drug in APL with potential to replace standard of care by addressing remaining unmet need
  • Encouraging PK Cross-Over Data:  Demonstrating oral bioavailability >90% and similar exposures between SY-2101 and IV ATO
  • Efficient Path to Approval: Comparable oral exposures to IV exposures in the Phase 1b study supports the potential for a smaller, more efficient path to approval

See Teaser linked below for information on the asset’s strengths, clinical information, and phase 3 readiness.
